New techniques for genetic improvement of horticultural brassicas. - HH0910SFV

Description
The objective of the project is to develop technology (DNA markers, mapping, doubled haploid production, clean gene transformation, embryo rescue) to increase the speed and efficiency of breeding vegetable brassicas. Resistance to some of the main UK brassica pests and diseases (Xanthomonas, spear rot, cabbage aphid, turnip mosaic virus) will be used as target characters to demonstrate the efficacy of the technology. Sources of resistance will be accessed from the Vegetable Gene Bank. Broccoli will be used as a model crop type where applicable. Material and information produced by the research will enable brassica breeders to provide UK growers with resistant varieties leading to a reduction in pesticide usage. The research programme addresses the policy and scientific objectives as given in the MAFF ROAME A for programme HH09 - Breeding for Annual Crops.
Objective
01. To develop and demonstrate the technology for interspecific introgression of genes of interest from the A (B rapa) and the B (B.nigra, B.fruticulosa) genome into the C genome (B.oleracea) using resistance to Xanthomonas campestris pv campestris, Brevicoryne brassica and turnip mosaic virus as target characters.

02. To determine the genetic control of responsiveness to microspore culture in B.oleracea.

03. To determine the effectiveness of reistance genes isolated from Arabidopis when deployed in B.oleracea by 'clean-gene' transformation using genes for resistance to Pseudomonas parasitica as target genes.

04. To determine the genetic control of resistance to spear rot caused by Pseudomonas.

05. To demonstrate differences in shelf life between broccoli breeding lines (for use in HH2121TFV - Long Life broccoli).
