Preparation of more sensitive bioassay models for the improved detection, differentiation & diagnosis of BSE. - SE1753

Description
At present, conventional mouse bioassay models represent the only feasible method for detection and characterisation of prion disease agents from cases of BSE and scrapie. However, mouse models are 1000 times less sensitive in detecting the agent than homologous bovine models.

This project aims to produce transgenic mice expressing prion proteins or chimaeric prion proteins from cattle or kudu, and then to challenge these mice with BSE agent to assess their susceptibility and sensitivity or resistance to disease.

Mouse oocytes will be injected with DAN constructs of bovine PrP whole ORF with 6 octarepeat element, kudu PrP whole ORF with 6 octarepeat element, kudu PrP whole ORF with 5 octarepeat element and bovine PrP bacterial artificial chromosome construct. The contractor is also looking at transgenic mice with ovine PrP genes which is funded by the EU.

01. Preparation of transgenic constructs.
02. Microinjections of transgenic DNA constructs.
03. Screening and analysis of transgenic mice.
04. Challenge of transgenic mouse lines with prion agent and analysis of mice and results.
05. Complete submission of reports.
