Interactions between orbiviruses and their hosts or vector species - SE2607

The work addresses MAFF's policy on the control of orbivirus-caused diseases by investigating the behaviour of the viruses in the vector Culicoides midge and the target mammalian species.

The effects of biological barriers to infection and the role of environmental factors, e.g. temperature, on virus replication in the midge will be examined.

The role of the BTV non-structural protein 3 in facilitating virus release will be investigated.

The determinants of cell attachment and entry in the vector and host species will be identified and compared.

The role of virus persistence in specific host cell types will be investigated as a mechanism of over-wintering of the virus.
1) To examine and identify barriers to the initial infection and subsequent dissemination of orbiviruses (BTV) in adult Culicoides, including particularly any barriers that may prevent the eventual release of virus into the salivary ducts and therefore vector status.

2) To examine the influence of environmental factors such as temperature on BTV infection rates and vector competence of adult Culicoides.

3) To examine the role of NS3 in the release of virus from infected insect and mammalian cells and the effect that such factors may have on the insect vector status.

4) To examine the processes of cell attachment and penetration by BTV core particles in mammalian and insect cells.

5) To examine the possible involvement of gamma delta T cells in over wintering mechanisms for orbiviruses.
