New strategies for vaccines against infectious bronchitis and the elucidation of pathogenicity and immunity - OD0712

Description
The subject of this proposal is the avian coronavirus infectious bronchitis virus (IBV) which infects chickens and pheasants. The overall technical objective is the production of IBV recombinants-IBVs containing genes from other IBV recombinants-IBVs containing genes from other IBV strains or marker genes. The scientific objective is to determine the molecular basis of pathogenicity/attenuation and cross-protective immunity. The strategic objective is to make possible the rational modification of IBV for vaccine purposes-to produce highly characterised, genetically stable vaccines in a short time and with the minimum of modification to pre-existing vaccines. This will stem both from the technical approaches that will be developed and the knowledge that will be gained from their application during the lifetime of the proposal.
Proposed modifications for vaccine design include: altering genes which determine pathogenicity/tropism i.e. to rationally attenuate IBV; swapping spike protein genes to change the protection-inducing range of an IBV strain, in response to changes in the field; tagging vaccine strains which will be of value for diagnostic and epidemiological purposes, including for regulatory authorities. The rapid development and timely application of required IBV vaccines will reduce the incidence of IB. A consequence of this will be a reduction in the application of antibiotics which are currently applied to control secondary bacterial infections which follow IBV infections. The proposed work will assist pharmaceutical companies to respond more rapidly and safely to the vaccine requirements of the poultry industry and to produce tagged vaccines to aid in problem solving during IB outbreaks.
