Novel Proteomic and biosensor based strategies for detection of downy mildew infection - HH3216SFV

Description
The overall aim is to identify protein biomarkers indicative of pea downy mildew (Peronospora viciae) and develop biosensors as a rapid and sensitive detection technology for this pathogen. This will be achieved by using proteomics to identify cellular proteins that are under- or over-expressed in pea plants with downy mildew infections. Proteins specific to downy mildew will be selected by comparison with protein expression in pea plants infected by other pathogens such as powdery mildew (Erysiphe pisi), grey mould (Botrytis cinerea) and bacterial blight (Pseudomonas syringe pv. pisi), and after wounding, an abiotic stess known to induce a common set of molecular responses. High affinity monoclonal antibodies will be raised against selected biomarkers. The marker proteins identified for P. viciae are likely to be produced by other Peronospora species. These cause diseases of field vegetable crops amounting to 80% of total planted area in the UK. It is therefore likely that a generic downy mildew detection system would be possible. Improved detection and diagnosis will provide information that can be applied within integrated management systems to minimise and improve efficiency of fungicide inputs and disease control. The final outcome will be antibiodies that can be applied in prototype devices for commercial exploitation. The proteomics data will also provide fundamental information for application in the development of novel control strategies for downy mildew diseases.
Objective
Project aim: To identify protein biomarkers that are indicative of pea downy mildew infection and investigate rapid detection technologies for this pathogen

Objective 1: To research and identify cellular biomarkers that are specifically over or under-expressed in pea plants infected by downy mildew.
Objective 2: To produce high affinity monoclonal antibodies (MAbs) to the specific biomarker(s) identified in WP1.
Objective 3: To write and present a coherent and constructive scientific report of the research undertaken.
