Immmunodetection of toxin Dinoflagellates - FC0715

R&D Summary Objectives:- Investigate the possibilities of developing a rapid test method for monitoring and detecting toxic algal species, based on immunodetection techniques. Key customer purpose:- Enable MAFF to better protect the public from consuming shellfish tainted with biotoxins by predicting the toxic potential of water regions, and determining the presence of potentially toxic organisms in shellfish.
To raise polyclonal antisera to both killed whole cell cultures and cell wall extracts of Alexandrium tamarense strains held by the Provasoli - Guillard centre for the culture of marine phytoplankton CCMP 115, 113, 1312, 117 and 118. A Gymnodinium aureolum strain from Sutton bay Plymouth and held at the MBA, Plymouth and amphidinium sp. and Prorocentrum sp. from British coasts held at the CCAP Oban. These antisera will then be evaluated and their specificity (between strains) ascertained by fluorescence microscopic examination, Ouchterlony gel immunoprecipitation, PAGE with Western blotting, and Continuous flow cytometry Electron microscopy and immunogold labelling will be performed if time allows) It is proposed that fluorescence microscopy and flow cytometric examination of natural samples probed with these antisera would detect and quantify (relatively) the distribution of these organisms within the natural environment.It is anticipated that new British strains of A. tamarense will be made available from the MAFF funded project studying these organisms led by Dr.J. Leftley. The toxin content of these strains will be ascertained at the University of Dundee under the supervision of Prof.G A. Codd.
